Billericay Town, Enfield Town and Hornchurch all had comfortable home wins, while Wingate & Finchley enjoyed success on the road as north London neighbours Haringey Borough earned a replay.
But Brentwood Town, Harrow Borough, Hendon and Woodford Town all bowed out after defeats.
Billericay Town romped to a 7-0 win over Stowmarket Town at New Lodge, with Ade Azeez opening the scoring from close range after just four minutes.
Bradley Stevenson added a second from the edge of the box, before Frankie Merrifield rattled the crossbar as the hosts went into the break two goals up.
Brooklyn Kabongolo added the third soon after the restart, with Azeez pouncing for his second just a minute later.
And Blues struck three more times in the final quarter of an hour, with Moses Emmanuel netting either side of Stevenson’s second.

Isthmian League Premier Division rivals Hornchurch scored five times in a 28-minute spell to see off Fakenham Town at Bridge Avenue.
Toby Byron and Lewwis Spence hit the woodwork as the scoreline remained blank in the first half, but Urchins opened the scoring on 56 minutes when Ade Yusuff netted from close range.
Substitute Sam Higgins struck on the hour, before Sean Scannell’s cross-shot from the right found the far corner to make it 3-0.
And Higgins fired in the fourth after Yusuff had dummied another Scannell cross from out wide, before Yusuff converted Tom Wraight’s centre to complete the nap hand for Steve Morison’s men on 84 minutes.
Enfield Town put out Potters Bar Town with a 3-0 win at the Queen Elizabeth Stadium.
Marcus Wyllie and Sam Youngs both struck inside the opening quarter of an hour to put Gavin Macpherson’s men on top.
And Joseph Payne made sure with the third goal midway through the second half.
Wingate & Finchley held on for a hard-fought 3-2 win over Whitstable Town, from two divisions lower, on the Kent coast.
Rhamar Garrett-Douglas made a goal-saving tackle at one end before setting up Anointed Chukwu to open the scoring on 38 minutes.
And Ruben Carvalho broke from midfield to made it 2-0 just before half-time.
The hosts hit back 15 minutes from time to raise their hopes, but Carvalho made it 3-1 with a 25-yard drive for his fifth of the season.
And although Whitstable scored again in stoppage time and had a late penalty appeal turned down, Wingate ensured their name would be in the hat for the draw.
Haringey Borough will also feature in Monday’s draw, after a late penalty earned them a 1-1 draw at Dulwich Hamlet.
Luke Wanadio gave Hamlet the lead from the spot on 17 minutes and it looked like being enough in front of a crowd of 1,714 at Champion Hill.
But Tom Loizou’s men were given a late lifeline and Matt Young stepped up to convert from 12 yards to keep them in the competition.

The two sides will meet again at Coles Park on Tuesday.
Brentwood Town were eight minutes away from a replay of their own, before Andrew Freeman’s own goal gave Hashtag United a 1-0 win at the Len Salmon Stadium.
And Southern League Premier outfit Hendon lost by the odd goal in five to Margate at Silver Jubilee Park, after taking a first-half lead through Hamza Semakula.
The visitors levelled through Sam Blackman just past the hour, with Fyn Rutherford putting them in front moments later.
Steve Cawley then made it 3-1, but Niko Muir replied for the Greens as four goals were scored in a 10-minute spell.
But Lee Allinson’s men could not get back on level terms in the time that remained.
Neighbours Harrow Borough were beaten 2-0 by Bracknell Town, meanwhile.
Joe Grant gave the visitors the lead with a 25-yard strike into the top corner on 28 minutes.
And George Franklin made sure in the last minute of normal time to send Jordan Berry’s men out.
Essex Senior League side Woodford Town saw their hopes of another upset shattered by Felixstowe & Walton United at Ashtons.
Billy Holland put the visitors in front midway through the first half, with Zak Brown and Joshua Hitter adding to the tally before the hour mark.
Nathan Sollosi hit back for Woods – who were unbeaten in 10 matches in all competitions – midway through the second half, but Noel Aitkens sealed a 4-1 win for in the last minute.
